Technically nope. There's one Prime-Canon example to the contrary: USS Yamato, NCC-1305-E. It is stated though, that this was a mistake done during production, done by a writer who wasn't fully aware of the limitations of the registration system. They retconned Yamato's registry onscreen lateron, three times actually. First to NCC-24383, which was only seen on a deployment list computer screen, then to NCC-71807, which is the definite and true registration and then there's the model of Yamato exploding, due to a computer malfunction caused by iconian software, which bears the registration NCC-71806.
On a technical level, the letter-suffix is only used for ships named Enterprise and only for ships that bear the name post Kirk's original 5-years mission. The reason being, that they want to honor this, for Starfleet and the Federation, ground-breaking 5-years mission.
There is one other canon-example though, but it's relevance can be debated due to the nature of the ship. It's the Wells-class USS Relativity, NCV-474439-G.
Also, there is the case of the second Defiant-class USS Defiant. She was supposed to be named Defiant-A, but for budgetary reasons they couldn't redo all the models and stock footage of the ship, so she never canonically got the suffix. As a rule of thumb, I suppose what remains is: If the ship is important enough to Starfleet-/Federation-history and is supposed to be a direct replacement of the ship bearing the name before, then the ship gets a suffix.
